Messenger RNA (mRNA) is an important platform for vaccines and future therapeutics. Its production is based on in vitro transcription (IVT), an enzymatic reaction in which nucleoside triphosphates (NTPs) are converted into RNA. To improve process control and efficiency, IVT can be operated in fed-batch mode, where NTPs are added during the reaction using a controlled feed.
In our process concept, the fed-batch IVT is monitored in real time using a mid-infrared (MIR) inline system. The measured data can be used to track mRNA formation and NTP consumption and to support mechanistic or data-driven models. After IVT, the process continues with purification steps.
The goal of this thesis is to connect the different process units, sensors, pumps, valves, analytics, and models into one automated workflow. A central part of the project is the development of a control system that enables automated data acquisition, process monitoring, and model-based process decisions. In the long term, this system should contribute to a digital mRNA manufacturing platform, where the process can be started once and then runs automatically from IVT to purified mRNA.
